ABSTRACT This study investigated the effect of calcium (Ca) and phytase interaction on growth performance and bone quality in 1-42-day-old broiler chickens. A total of 624 female one-day-old Ross 308 broilers were allotted to 13 treatments with four replicates and 12 birds per replicate. A 2 × 6 factorial experiment was designed to test the combinations of 0.50% and 1.00% Ca with 0, 500, 1,000, 2,500, 5,000, and 10,000 FTU/kg phytase in the basal diet (0.25% non-phytate phosphorus, NPP). The control diet contained adequate Ca and phosphorus (P). Dietary Ca, phytase, and their interaction affected growth performance and bone mineralization of broilers at 1-42 days of age (p<0.05). The broilers fed with 1.00% Ca had lower body weight gain (BWG) and feed intake (FI) compared with the birds fed with 0.50% Ca (p<0.05). The BWG, FI, leg bone weight, and ash weight of the broilers fed with 0.25% NPP were lower than those of birds fed with the control diet (p<0.05). The addition of 500-10,000 FTU/kg phytase improved growth rate and leg bone quality, especially at 1.00% Ca (p<0.05). No differences were observed in growth performance and bone quality of 42-day-old broilers fed with 1.00% Ca + 2,500-10,000 FTU/kg phytase and the control diet (p>0.05). These data indicated that high doses of phytase (2,500-10,000 FTU/kg) alleviate the negative effects of Ca and P imbalance (Ca-to-NPP ratio = 4.0) on growth performance and bone mineralization of broiler chickens.

The properties and deterioration in strength of cemented tailings backfill (CTB )in the underground acidic environment under oxidizing conditions were studied. X-ray diffraction analysis and scanning electron microscopy of the surface of the CTB was carried out, and the sulphuric acid corrosion mechanism elucidated. The properties tested included compressive strength, elastic modulus, cohesion, internal friction angle, variation in the hydrogen ion concentration, and stress-strain relationship in different corrosion periods. The damage model of the CTB was established considering the effects of parameters such as corrosion time and strain on the damage evolution. It was found that the compressive strength, elastic modulus, cohesion (binder effect), and internal friction angle increased at first and then decreased with exposure time. In the long term, the peak stress decreased with corrosion time while the peak strain increased; elastic modulus, and deformation modulus also decreased. The damage caused by corrosion and by load were related by means of a mathematical model, which revealed the relationships between sulphuric attack, load, and damage to backfill in complex underground environments. During corrosion, the expansion of gypsum and ettringite caused microfractures in the CTB. With increasing corrosion time, micro-cracks developed and proliferated. The mechanism of corrosion damage was found to be the dissolution of hydrogen ions and a sulphate ion reaction that produces an expansive substance, resulting in deterioration of the strength of CTB.

ABSTRACT Homeobox B9 (HOXB9) gene has been demonstrated to be associated with melanogenesis in chicken plumage by high-throughput RNA sequencing. In this study, we cloned and characterised HOXB9 in black-boned chickens. Two alternative splice variants (HOXB9-1 and HOXB9-2) were identified in chicken feather bulbs. Expression analysis of HOXB9 in 11 different chicken tissues by RT-PCR indicated that the two transcripts were only expressed in the kidney, abdominal fat, feather bulbs, skin, and small intestine. No HOXB9-1 or HOXB9-2 transcripts were detected in the breast muscle or the ovary. The two HOXB9 variants were expressed at significantly different levels in black feather bulbs and white feather bulbs (p<0.01), and in black skin compared with white skin (p<0.01). The results indicate that HOXB9-1 and HOXB9-2 may be involved in melanin formation in the plumage and skin. The expression of HOXB9-2 was higher in white than in black muscles (p<0.05), HOXB9-2 may play a role in muscle melanogenesis. Our results suggest that HOXB9, which is partially regulated by alternative splicing, may be involved in the process of melanogenesis in the black-boned chicken.

Recent studies have revealed that an intrinsic apoptotic signaling cascade is involved in vascular hyperpermeability and endothelial barrier dysfunction. Propofol (2,6-diisopropylphenol) has also been reported to inhibit apoptotic signaling by regulating mitochondrial permeability transition pore (mPTP) opening and caspase-3 activation. Here, we investigated whether propofol could alleviate burn serum-induced endothelial hyperpermeability through the inhibition of the intrinsic apoptotic signaling cascade. Rat lung microvascular endothelial cells (RLMVECs) were pretreated with propofol at various concentrations, followed by stimulation with burn serum, obtained from burn-injury rats. Monolayer permeability was determined by transendothelial electrical resistance. Mitochondrial release of cytochrome C was measured by ELISA. Bax and Bcl-2 expression and mitochondrial release of second mitochondrial-derived activator of caspases (smac) were detected by Western blotting. Caspase-3 activity was assessed by fluorometric assay; mitochondrial membrane potential (Δψm) was determined with JC-1 (a potential-sensitive fluorescent dye). Intracellular ATP content was assayed using a commercial kit, and reactive oxygen species (ROS) were measured by dichlorodihydrofluorescein diacetate (DCFH-DA). Burn serum significantly increased monolayer permeability (P<0.05), and this effect could be inhibited by propofol (P<0.05). Compared with a sham treatment group, intrinsic apoptotic signaling activation - indicated by Bax overexpression, Bcl-2 downregulation, Δψm reduction, decreased intracellular ATP level, increased cytosolic cytochrome C and smac, and caspase-3 activation - was observed in the vehicle group. Propofol not only attenuated these alterations (P<0.05 for all), but also significantly decreased burn-induced ROS production (P<0.05). Propofol attenuated burn-induced RLMVEC monolayer hyperpermeability by regulating the intrinsic apoptotic signaling pathway.

With the increasingly acute contradictions between shortage of bonanza phosphorus resources and phosphate fertilizer demands, to develop an efficient use technology for low-grade phosphate rocks (LGPR) was an inevitable choice. In this study, the physicochemical pro-release phosphate rock (PCPR) was prepared by adding modified lignin during the process of ultra-fine grinding LGPR. The relationship between phosphorus chemical composition of PCPR, molecular structure and fertilizer efficiency was explored by chemical analysis, spectral analysis and pot experiment. The results showed that after pro-release treatment of phosphate rock (PR), available phosphorus, reactive phosphorus and soluble phosphorus levels were significantly increased compared with PR. The available phosphorus content increased by 40%, the reactive phosphorus content increased more than 3.5 times, and 6 consecutive extraction soluble phosphorus accumulation increased by 24 times. The structure of the PR showed the pro-release processing would significantly increase particle fineness and specific surface area of PR , strengthen the characteristic absorption peak intensity of PO4(3-), HPO4(2-), replace the Cl-, F-, Fe²+ in the apatite and change the phase composition of PR. Pot experiment results showed corn biomass and phosphorus utilization processed by the PCPR were much higher than superphosphate (SP), diammonium phosphate(DAP), and significantly improve soil phosphorus content. The high soluble phosphorus and active phosphorus in PCPR providing the moderate continuous phosphorus supply intensity was the main reason for its high bioavailability. The combination of a static extraction and continuous extraction of reactive phosphorus and soluble phosphorus was an appropriate evaluation of physicochemical pro-release effect.

Considering the effects of the changes of the pore water pressure around the opening, the construction sequence of the tunnel and the interaction between the liner and the surrounding geomaterial on the mechanical response of the tunnel and in conjunction with the analyses of the continuity and boundary conditions for the stress and displacement and hydraulic conditions, the elastic problem for a deep pressure tunnel with impermeable liner in a saturated elastic porous media that obeys Terzaghi's effective stress principle is investigated. The influences of the relative liner thickness and rigidity and the relative distance of the point under investigation to the tunnel axis on the stress-displacement fields for various combinations of the mechanical and geometric parameters are evaluated and discussed.

Myocardial ischemic preconditioning up-regulated protein 1 (Mipu1), a novel zinc finger protein, was originally cloned using bioinformatic analysis and 5' RACE technology of rat heart after a transient myocardial ischemia/reperfusion procedure in our laboratory. In order to investigate the functions of Mipu1, the recombinant prokaryotic expression vector pQE31-Mipu1 was constructed and transformed into Escherichia coli M15(pREP4), and Mipu1-6His fusion protein was expressed and purified. The identity of the purified protein was confirmed by mass spectrometry. The molecular mass of the Mipu1 protein was 70.03779 kDa. The fusion protein was intracutaneously injected to immunize New Zealand rabbits to produce a polyclonal antibody. The antibody titer was approximately 1:16,000. The antibody was tested by Western blotting for specificity and sensitivity. Using the antibody, it was found that Mipu1 was highly expressed in the heart and brain of rats and was localized in the nucleus of H9c2 myogenic cells. The present study lays the foundation for further study of the biological functions of Mipu1.

Os capins híbridos obtidos pelo cruzamento entre Sorghum sudanensis (capim-sudão) e genótipos de Sorghum bicolor possuem alto potencial para produção de biomassa e para controle de plantas daninhas pela produção de substâncias fitotóxicas, como o sorgoleone (uma hidroquinona de cadeia longa). Sementes de alface (Lactuca sativa), tomate (Lycopersicum sculentum), beldroega (Portulaca oleracea) e caruru (Amaranthus retroflexus) foram submetidas a tratamentos com extratos aquosos da parte aérea e das raízes do híbrido de sorgo com capim-sudão, cv. Trudan 8, colhido em cinco diferentes estádios de crescimento (10, 20, 30, 40 e 50 dias após a emergência). Os extratos foram preparados em quatro concentrações (0, 0,4, 2 e 10 g L-1, p/v) e aplicados em quatro repetições. Após os tratamentos, a germinação e o comprimento de plântulas das espécies foram avaliados. A germinação de sementes de tomate, caruru e alface foi inibida pelos extratos da parte aérea das plantas de Trudan 8, na concentração de 10 g L ¹, colhidas até os 20 dias após a emergência. A germinação de sementes de beldroega, no tocante à porcentagem de germinação, não foi inibida pelos extratos de Trudan 8. O crescimento das quatro espécies avaliadas foi inibido quando tratadas com extratos aquosos da parte aérea de Trudan 8, colhida até os 10 dias após a emergência, na concentração de 10 g L-1.

Hybrids of Sorghum sudanensis (sudangrass) and Sorghum bicolor genotypes can produce high amounts of biomass, sorgoleone (a long chain hydroquinone), and other phytotoxic substances. Shoots and roots of a sorghum-sudangrass hybrid (cv. Trudan 8) were collected 10, 20, 30, 40, and 50 days after emergence. Four concentrations of aqueous extracts from the shoots and roots (0, 0.4, 2, and 10 g L-1, w/v) were used to treat seeds of lettuce (Lactuca sativa), tomato (Lycopersicum sculentum), purslane (Portulaca oleracea), and pigweed (Amaranthus retroflexus). Seed germination of lettuce, tomato, and pigweed was inhibited by extracts from sorghum-sudangrass shoots at 10 g L-1 when made from sorghum-sudangrass plants 20 days or less in age. Seed germination of purslane was not inhibited by any sorghum-sudangrass extract. Growth of the four species evaluated were systematically inhibited when treated with 10 g L-1 extracts from sorghum-sudangrass shoots harvested up to 10 days after emergence.

The Chinese Erhualian is one of the most prolific pig breeds in the world, but it is in danger of being replaced by other exotic pig breeds because of its slow growth rate and high fat content in the body. To obtain some genetic information for conservation, we analysed the Erhualian pigs by using a PCR-RFLP for the calcium-release-channel (CRC) gene, nine polymorphic microsatellites and the complete mtDNA D-loop sequences, and compared these data with those from other pig breeds from Europe and Asian. The PCR-RFLP analysis of the CRC gene showed that the frequency of the C allele associated with stress resistance was 100% in the Erhualian pigs. Neighbour-Joining trees constructed on the basis of mtDNA D-loop sequences and the microsatellite analysis clearly showed that the Erhualian pigs were located in a separate branch. These data suggest that the Erhualian pigs are different from other breeds. Microsatellite analysis showed that the average allele number (5.3/locus) in the Erhualian pig was intermediate as compared with that (4.8-7.0/locus) in the three European pig breeds. The expected heterozygosity was higher in the Erhualian pig (0.78) than that in these European pig breeds (0.59-0.72), whereas the observed heterozygosity was higher in the European breeds (0.51-0.64) than in the Erhualian pig (0.46). In the Erhualian pig, the fixation index (F IS) was as high as 0.41. These data suggest a high level of inbreeding and/or subpopulation in the Erhualian pigs. For conservation of the germplasm in the Erhualian pigs, it is necessary to take measures to reduce inbreeding and/or subpopulation.
